Abstract

Kulit buah jeruk sambal (Citrus spp.) diketahui mengandung senyawa fenolik dan flavonoid yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk menentukan profil kromatografi lapis tipis (KLT) dan nilai Rf senyawa fenolik serta flavonoid pada fraksi metanol kulit buah jeruk sambal. Sampel kulit buah dikeringkan, dihaluskan, kemudian diekstraksi dengan etanol dan difraksinasi menggunakan metanol. Uji KLT dilakukan pada pelat silika gel G60 F254 dengan fase gerak metanol:etil asetat dalam berbagai perbandingan. Identifikasi bercak menggunakan pereaksi FeCl₃ 10% untuk fenolik dan AlCl₃ 5% untuk flavonoid, kemudian diamati di bawah sinar UV 254 nm dan 366 nm. Hasil menunjukkan bahwa senyawa fenolik terdeteksi dengan nilai Rf standar 0,73 pada eluen Metanol:Etil Asetat = 8:2 dan 0,58 pada eluen Metanol:Etil Asetat = 10:0. Senyawa flavonoid memiliki nilai Rf standar 0,74 pada eluen Metanol:Etil Asetat = 0:10, 0,75 pada eluen Metanol:Etil Asetat = 2:8, 0,76 pada eluen Metanol:Etil Asetat = 7:3, dan 0,78 pada eluen Metanol:Etil Asetat = 10:0. Kesimpulan penelitian ini menyatakan bahwa fraksi metanol kulit buah jeruk sambal positif mengandung fenolik dan flavonoid dengan nilai Rf dalam rentang standar 0,2–0,8, sehingga berpotensi dikembangkan sebagai sumber bahan aktif alami.

Abstract (English)

The peel of Citrus spp. is known to contain phenolic and flavonoid compounds with antioxidant potential. This study aimed to determine the thin layer chromatography (TLC) profile and Rf values of phenolic and flavonoid compounds in the methanol fraction of Citrus spp peel. The peel samples were dried, powdered, extracted with ethanol, and fractionated using methanol. TLC was performed on silica gel G60 F254 plates with methanol:ethyl acetate as the mobile phase in different ratios. Spots were identified using 10% FeCl₃ for phenolics and 5% AlCl₃ for flavonoids, observed under UV light at 254 nm and 366 nm. The results showed that phenolic compounds exhibited Rf values of 0.73 at eluent Methanol:Ethyl Acetate = 8:2 and 0.58 at Methanol:Ethyl Acetate = 10:0. Flavonoid compounds presented Rf values of 0.74 at Methanol:Ethyl Acetate = 0:10, 0.75 at Methanol:Ethyl Acetate = 2:8, 0.76 at Methanol:Ethyl Acetate = 7:3, and 0.78 at Methanol:Ethyl Acetate = 10:0. The study concludes that the methanol fraction of Citrus spp. peel contains phenolic and flavonoid compounds with Rf values within the standard range of 0.2–0.8, indicating its potential as a natural source of bioactive compounds for pharmaceutical or cosmetic applications.
