Abstract

Daun kratom (Mitragyna speciosa Korth) merupakan tanaman tradisional yang mengandung senyawa alkaloid dengan potensi farmakologis. Penelitian ini bertujuan untuk mengidentifikasi kandungan alkaloid pada fraksi etanol dan n-heksana daun kratom, dan menentukan kadar alkaloid total kedua fraksi tersebut. Simplisia daun kratom diekstraksi dengan maserasi etanol 96%, kemudian difraksinasi menggunakan n-heksana dan etanol. Uji kualitatif alkaloid dilakukan dengan pereaksi Mayer, Wagner, dan Dragendorff, sedangkan kadar alkaloid total diukur secara spektrofotometri UV-Vis pada λ 273 nm menggunakan kafein sebagai standar. Hasil uji kualitatif menunjukkan adanya alkaloid pada kedua fraksi, ditandai dengan pembentukan endapan putih (Mayer), cokelat (Wagner), dan merah bata (Dragendorff). Analisis kuantitatif menunjukkan kadar alkaloid total fraksi n-heksana (80,565%) lebih tinggi dibandingkan fraksi etanol (20,858%). Temuan ini menunjukkan bahwa pelarut non-polar (n-heksana) lebih efektif mengekstrak alkaloid daripada pelarut polar (etanol).

Abstract (English)

Mitragyna speciosa Korth Leaves contain bioactive alkaloids with pharmacological potential. This study aimed to identify alkaloids in ethanol and nhexane fractions of kratom leaves, and determine the total alkaloid content of both fractions. Dried kratom leaves were macerated with 96% ethanol, then fractionated using n-hexane and ethanol. Qualitative alkaloid tests were performed with Mayer’s,Wagner’s, and Dragendorff’s reagents, while total alkaloid content was measured by UV-Vis spectrophotometry at λ 273 nm using caffeine as a standard. Qualitative tests confirmed alkaloids in both fractions, indicated by white (Mayer), brown (Wagner), and brick-red (Dragendorff) precipitates. Quantitative analysis showed the n-hexane fraction had significantly higher total alkaloid content (80.565%) and ethanol fraction (20.858%), showing non-polar solvents are more effective for alkaloid extraction
