Abstract

Escherichia coli adalah mikroba penyebab infeksi yang paling sering dijumpai sehari-hari dan menghasilkan senyawa toksin yang berbahaya bagi manusia. Dalam mengatasi masalah tersebut diperlukan upaya pengendalian kehidupan bakteri penyebab luka infeksi. Penelitian ini bertujuan untuk menguji aktivitas antibakteri dari sediaan gel nanokoloid perak hasil biosintesis menggunakan ekstrak tanaman keladi sarawak dan AgNO3 dengan variasi 0,05 M, 0,10 M, dan 0,15 M terhadap bakteri Escherichia coli. Sediaan gel nanokoloid perak ini dibuat dengan basis gel yang bersifat hidrofilik karena memiliki keunggulan seperti stabilitas nya lebih besar, daya sebarnya pada kulit baik, mudah dicuci dengan air dan pelepasan obat baik sehingga meningkatkan efektivitas penggunaan gel sebagai antibakteri. Uji aktivitas antibakteri dilakukan dengan metode difusi menggunakan kertas cakram dan dilakukan pengukuran zona bening yang terbentuk di sekitar kertas cakram. Hasil pengukuran rata-rata diameter daya hambat yaitu Formula I (0,05 M) sebesar 10,84 mm, Formula II (0,10 M) sebesar 12,67 mm, Formula III (0,15 M) sebesar 14,97 mm dan kontrol positif sebesar 24,66 mm. Berdasarkan hasil penelitian tersebut dapat disimpulkan bahwa sediaan gel nanokoloid perak hasil biosintesis menggunakan ekstrak tanaman keladi sarawak (Alocasia macrorrhizos(L.)) memiliki daya hambat terhadap bakteri Escherichia coli. Dan berdasarkan hasil statistik uji ANOVA One Way Post-Hoc test menunjukkan bahwa nilai dari F1,F3 dan kontrol positif memiliki nilai signifikasi <0,05 yang berarti perlakuan tersebut memiliki perbedaan yang nyata. Sedangkan F2 memiliki nilai signifikasi ≥ 0,05 sehingga dapat disimpulkan bahwa tidak terdapat perbedaan yang nyata antara F2 dengan ketiga perlakuan tersebut dalam menghambat bakteri Escherichia coli.

Abstract (English)

Escherichia coli is the most common microbe that causes daily infections and produces toxin compounds that are harmful to humans. In overcoming this problem, efforts are needed to control the life of bacteria that cause wound infections. This study aims to test the antibacterial activity of biosynthesized silver nanocolloid gel preparations using Sarawak taro plant extract and AgNO3 with variations of 0.05 M, 0.10 M, and 0.15 M against Escherichia coli bacteria. This silver nanocolloid gel preparation was made with a hydrophilic gel base because it has advantages such as greater stability, good skin spreadability, easy washing with water and good drug release so as to increase the effectiveness of using the gel as an antibacterial. The antibacterial activity test was carried out by diffusion method using disc paper and measuring the clear zone formed around the disc paper. The results of measuring the average diameter of the inhibitory power are Formula I (0.05 M) by 10.84 mm, Formula II (0.10 M) by 12.67 mm, Formula III (0.15 M) by 14.97 mm and positive control by 24.66 mm. Based on the results of this study, it can be concluded that the biosynthesized silver nanocolloid gel preparation using Sarawak taro plant extract (Alocasia macrorrhizos(L.)) has inhibition against Escherichia coli bacteria. And based on the statistical results of the ANOVA One Way Post-Hoc test, it shows that the values of F1, F3 and positive control have a significance value <0.05, which means that the treatment has a significant difference. While F2 has a significance value ≥ 0.05 so it can be concluded that there is no significant difference between F2 and the three treatments in inhibiting Escherichia coli bacteria
