Abstract

Nanopartikel perak (NPP) diketahui memiliki aktivitas antibakteri dan sering dimanfaatkan di bidang kesehatan sebagai antimikroba yang dapat diaplikasikan terhadap luka. NPP dapat disintesis secara ramah lingkungan menggunakan bahan alami seperti ekstrak kulit jeruk kunci (Citrus microcarpa Bunge) yang mengandung flavonoid dan senyawa bioaktif lainnya. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari NPP hasil biosintesis menggunakan ekstrak kulit jeruk kunci terhadap bakteri Escherichia coli. Sintesis dilakukan dengan mencampurkan larutan AgNO₃ 0,15 M dengan ekstrak pada konsentrasi 10%, 15%, dan 20%. Pengujian dilakukan menggunakan metode difusi cakram dengan Neomycin sulfate sebagai kontrol positif. Hasil menunjukkan bahwa seluruh konsentrasi NPP mampu menghambat pertumbuhan Escherichia coli, ditandai dengan terbentuknya zona bening. Rata-rata diameter zona hambat yang dihasilkan sebesar 7,88 mm (10%), 8,76 mm (15%), 12,12 mm (20%) dan 22,37 mm (kontrol positif), rata-rata tertinggi ditemukan pada konsentrasi 20% yaitu sebesar 12,12 mm dan termasuk kategori kuat. Uji ANOVA menunjukkan adanya perbedaan signifikan antar perlakuan (p < 0,05), namun uji post hoc Bonferroni menunjukkan tidak ada perbedaan signifikan antar ketiga konsentrasi uji. Meskipun begitu, secara deskriptif, konsentrasi 10% menunjukkan daya hambat terbaik. Hasil ini menunjukkan potensi NPP dari ekstrak kulit jeruk kunci sebagai agen antibakteri terhadap Escherichia coli.

Abstract (English)

Silver nanoparticles (NPP) are known to have antibacterial activity and are often used in the health sector as antimicrobials that can be applied to wounds. NPP can be synthesized in an environmentally friendly manner using natural ingredients such as key orange peel extract (Citrus microcarpa Bunge) which contains flavonoids and other bioactive compounds. This study aims to determine the antibacterial activity of NPP biosynthesized using key orange peel extract against Escherichia coli bacteria. Synthesis was carried out by mixing 0.15 M AgNO₃ solution with extracts at concentrations of 10%, 15%, and 20%. Testing was carried out using the disc diffusion method with Neomycin sulfate as a positive control. The results showed that all concentrations of NPP were able to inhibit the growth of Escherichia coli, indicated by the formation of a clear zone. The average diameter of the inhibition zone produced was 7.88 mm (10%), 8.76 mm (15%), 12.12 mm (20%) and 22.37 mm (positive control), the highest average was found at a concentration of 20%, which was 12.12 mm and was included in the strong category. The ANOVA test showed a significant difference between treatments (p <0.05), but the Bonferroni post hoc test showed no significant difference between the three test concentrations. However, descriptively, the 10% concentration showed the best inhibition. These results indicate the potential of NPP from Citrus microcarpa Bunge extract as an antibacterial agent against Escherichia coli.
